Sermorelin is a growth hormone–releasing hormone (GHRH) analog — the biologically active 1–29 fragment of human GHRH, the shortest piece that keeps full activity. Rather than supplying growth hormone, it prompts the pituitary to make and release the body’s own.
How sermorelin works
Sermorelin binds the GHRH receptor on the anterior pituitary and stimulates the gland to synthesise and secrete growth hormone (GH). Because that release remains under the body’s normal feedback controls (via somatostatin), sermorelin tends to produce a more physiologic, pulsatile GH pattern than injecting GH directly. Downstream, GH raises IGF-1, the mediator of many of its tissue effects.
What the research shows
- GH-deficiency diagnosis & pediatric treatment (Prakash & Goa, 1999). A review documenting sermorelin’s dual role — a diagnostic test of GH reserve, and a treatment that induced catch-up growth in most GH-deficient children.
- Stimulates GH release (Wilton et al., 1993). GHRH(1-29) stimulated GH secretion in healthy subjects after IV or intranasal dosing.
- Adult GH-deficiency testing (Biller et al., 2002). The arginine + GHRH test matched the insulin-tolerance test for diagnosing adult GH deficiency, with high sensitivity and specificity.
- Nightly GHRH in older men (Vittone et al., 1997). Nightly GHRH raised nocturnal GH and improved some strength measures, though it did not significantly raise IGF-1.
Historical note: sermorelin was originally developed as a diagnostic and treatment agent for growth-hormone deficiency (marketed as Geref) and was later discontinued for commercial — not safety or efficacy — reasons. Candidly, its strongest evidence is in GH-deficiency diagnosis and pediatric treatment; data in healthy aging adults are older and limited. Use belongs under a physician’s guidance.
