AOD-9604 is a small synthetic fragment of human growth hormone — specifically the C-terminal “lipolytic” tail (residues 176–191) with an extra tyrosine. It was designed to capture GH’s fat-burning action without its growth or blood-sugar effects.
How AOD-9604 works
AOD-9604 is the isolated fat-metabolising domain of human growth hormone. In research it reproduces GH’s stimulation of lipolysis (fat breakdown) and fat oxidation, and up-regulates β3-adrenergic receptor expression in fat tissue — but it does so without acting through the GH receptor, and human studies found no rise in IGF-1. That separation from the growth/IGF-1 axis is the whole point of the molecule.
What the research shows
- Fat metabolism in animals (Ng et al., 2000). Oral AOD-9604 cut body-weight gain by more than half in obese rats and raised fat-tissue lipolytic activity without impairing insulin sensitivity.
- Fat oxidation (Heffernan et al., 2001). Chronic AOD-9604 increased fat oxidation and reduced weight in obese mice, mirroring GH’s lipolytic action.
- Human safety (Stier et al., 2013). Across six randomised placebo-controlled trials, AOD-9604 was well tolerated with a safety profile like placebo and no effect on IGF-1.
Candour: despite promising animal data, the larger human obesity trials did not show significant weight loss versus placebo, and the original anti-obesity drug program was discontinued. Use belongs under a physician’s guidance.
